Protein architecture is the key to gene function. The specific amino acid sequence determines the general shape, binding properties, and reactivity of the protein.
Mutations in DNA result in changes in the sequence of amino acids of a protein (its primary structure). Thus, mutations may lead to changes in the way a protein functions and can become the molecular cause of Sickle Cell Anemia
Phenylketonuria (PKU) is a type of amino acid metabolism disorder. It is very rare, inherited in most cases. If you have it, your body can’t process part of a protein called phenylalanine (Phe). Phe is in almost all foods. If your Phe level gets too high, it can damage your brain and cause severe intellectual disability. All babies born in U.S. hospitals must now have a screening test for PKU. This makes it easier to diagnose and treat the problem early.
NOTE: Adults can develop PKU after 40, mostly after enzyme or hormonal disruption caused by emotional or physical stress and close to menopause
PKU is caused by mutations in the gene that helps make an enzyme called phenylalanine hydroxylase (pronounced fen-l-AL-uh-neen hahy-DROK-suh-leys), or PAH. This enzyme is needed to convert the amino acid phenylalanine into other substances the body needs. When this gene, known as the PAH gene, is defective, the body cannot break down phenylalanine.
Amino acids help build protein, but phenylalanine can cause harm when it builds up in a person’s body. In particular, nerve cells in the brain are sensitive to phenylalanine.
Protein is an important nutrient that is key to every cell in your body. Your body needs protein to build and repair tissues — and to make enzymes and hormones. Healthy bones, blood, hair, nails and muscles all rely on protein and since our bodies can’t store it, we must get high quality protein from our food.
